Terrific Broth Media

  1. Add the following to 800 mL distilled H2O: 12 g Tryptone, 24 g yeast extract, and 4 mL glycerol.

  2. Adjust to 900 mL with distilled H2O.

  3. Sterilize by autoclaving.

  4. Allow to cool to room temperature.

  5. Adjust volume to 1,000 mL with 100 mL of a filter sterilized solution of 0.17 M KH2PO4 and 0.72 M K2HPO4.
