Cross-linking with BS3

BS3 (bis(sulfosuccinimidyl) suberate)

Procedure

  1. Added to the sample buffer at a concentration of 0.25 mM.

  2. Incubate the mixture on ice for 2 hours before quenching with 10 mM Tris-Cl.

  3. The cross-linked sample will be characterized using SDS-PAGE, SEC, and negative-stain EM.

Notes

  1. The primary amine of the lysine side chain and the N-terminus of each polypeptide are available as targets for NHS-ester crosslinking reagents.

  2. The soluble up to ~100 mM in water.
